What is Rybelsus?

Rybelsus® is a once-daily oral tablet containing semaglutide — the same active ingredient as Ozempic and Wegovy, but in a pill rather than an injection. It is the only oral GLP-1 receptor agonist currently available and represents an important option for patients who are needle-averse or who strongly prefer not to inject.

It is approved for Type 2 Diabetes and is used for weight management under medical supervision.


How does Rybelsus work?

Oral semaglutide works through the same GLP-1 receptor mechanism as injectable semaglutide:

  • Reduces appetite by signalling fullness to the brain
  • Slows gastric emptying to extend satiety after meals
  • Stimulates insulin release in response to elevated blood sugar
  • Reduces glucose production from the liver

Because the oral formulation has lower bioavailability than the injectable version — meaning less of the active ingredient reaches the bloodstream — the weight loss effect is somewhat less pronounced than Ozempic or Wegovy. However for patients who won’t inject, it remains a clinically meaningful option.


Who is Rybelsus for?

Rybelsus may be suitable for:

Rybelsus dosing

Rybelsus is available in three doses and titrated over time.

  • Month 1: 3mg once daily (introductory dose)
  • Month 2: 7mg once daily
  • Month 3 onwards: 14mg once daily if needed

Important administration requirements — Rybelsus must be taken on an empty stomach, with no more than 120ml of plain water. No food, drink (other than water), or other medications for at least 30 minutes after taking it. This is not optional — food and other medications significantly reduce how much semaglutide is absorbed.


What results can you expect?

Clinical trials of oral semaglutide at 14mg show average weight loss of approximately 5–10% of body weight over 26–52 weeks when combined with lifestyle changes. This is less than injectable semaglutide (Ozempic/Wegovy) due to lower bioavailability, but meaningful for patients for whom injections are not an option.

For patients with Type 2 Diabetes, Rybelsus also produces significant improvements in HbA1c and fasting blood glucose.

Possible side effects

Common side effects, particularly at the start of treatment:

  • Nausea
  • Vomiting
  • Diarrhea or constipation
  • Abdominal discomfort

These are similar to injectable GLP-1 medications and typically improve as your body adjusts. The empty-stomach administration requirement can itself cause mild nausea in some patients — your doctor will advise on managing this.
